Bile acids react with thio-NAD under the catalysis of 3α -hydroxysteroid dehydrogenase (3α-HSD) to generate thio-NADH and 3α -ketosteroids. The latter reacts with NADH to generate bile acids and NAD under the action of 3α-HSD enzymes. Within a certain reaction time, the thio-NADH produced by the enzyme cycle is proportional to the concentration of bile acids in the sample. Based on this, the content of total bile acids can be calculated.
