ssK36 is an enhanced substrate peptide for histone methyltransferase (SET) domain protein 2 (SETD2). ssK36 was specifically designed for SETD2, a particular PKMT responsible for adding a methyl group to the 36th lysine residue of histone H3 (H3K36) in human cells, forming H3K36me3. The methylation rate of ssK36 is more than 100 times faster than that of the natural substrate H3K36. ssK36 is used to study the catalytic mechanisms of PKMTs, especially substrate specificity and catalytic efficiency.
