SAP15 (Synthetic anti-inflammatory peptide 15) is a synthetic anti-inflammatory peptide composed of 15 amino acids, designed from human β-defensin 3. This peptide possesses the ability to penetrate cells and induces a downregulation of intracellular inflammatory responses. It inhibits inflammatory responses by reducing NF-κBp65 phosphorylation by inhibiting HDAC5 phosphorylation. In LPS-induced macrophages, SAP15 effectively inhibits the phosphorylation of both HDAC5 and NF-κBp65. Furthermore, SAP15 treatment promotes the expression of aggrecan and type II collagen in LPS-induced chondrocytes and reduces osteocalcin expression. SAP15 is suitable for research on inflammation modulation and anti-inflammatory therapy in biomaterials.
