Recombinant CRISPR-Cas14a1 protein

Cas14a1 nuclease is a type of endonuclease. Guided by tracrRNA:crRNA or sgRNA， it can specifically bind and cleave target single-stranded DNA without requiring a PAM site. The molecular weight of our Cas14a1 is only 61.9 kDa. Similar to Cas12， Cas14a1 can bind to target nucleic acids and activate its trans-cleavage activity towards ssDNA， enabling its application in molecular detection of target nucleic acids. Unlike Cas12， Cas14a1 only binds to ssDNA targets. Therefore， amplified and enriched target nucleic acids need to be treated with T7 exonuclease， and one of the amplification primers should be modified with phosphorothioation to ensure that T7 exonuclease only cleaves one strand， leaving the ssDNA target strand for Cas14a1-mediated molecular detection.
