AatII activity is inhibited at low ionic strength. Enzyme activity decreases if the reaction buffer pH is not between 7.5 and 8.0 at 25°C; cleavage may be inhibited for CpG-methylated DNA; aster activity was not observed after 3 hours of incubation, but may appear after longer digestion; Inactivation conditions: 80°C for 20 minutes. QuickCut fast restriction enzymes are a series of genetically engineered rapid endonucleases, suitable for the rapid digestion of plasmid DNA, PCR products, genomic DNA and other templates. All QuickCut fast restriction enzymes exhibit high activity in both Reaction Buffer and Reaction Color Buffer, and complete DNA digestion within 5–15 minutes. Reaction Color Buffer contains red and yellow tracking dyes, allowing the digested products to be directly loaded for gel electrophoresis. The red dye migrates at a rate similar to a 2500 bp double-stranded DNA fragment in 1% agarose gel; the yellow dye migrates at a rate similar to a 10 bp double-stranded DNA fragment in 1% agarose gel.
