Measurement Principle: Reducing agent will make the disulfide bond cleaved, the cleaved sulfhydryl group will undergo nucleophilic reaction, that is, the sulfhydryl group reacts with 5,5'-dithio-bis-nitrobenzoic acid (DTNB) to produce a yellow color compound, with a maximum absorption peak at 412 nm, according to which, we can calculate the content of the disulfide bond of the protein.
