The product is stable at ≤-20℃ and is used in aliquots to minimize the number of freeze-thaw cycles. Please do not store in a frost-free refrigerator, and be sure to keep the product in a frozen state when storing it for a long time. When thawing the product for the first time, encapsulate the vial in crushed ice and place it in a 4℃ freezer to allow it to thaw. For all consumables that come into contact with the product, please cool down in advance. Please do not hold the container containing this product with your hands for too long during use to prevent body temperature from making the product gel; If the product is thick and viscous in a short period of time, you can re-place the product in an environment of 0℃-4℃ for 1-2 hours to restore fluidity without affecting use. The experimental operator needs to strictly distinguish the experimental operation table, the clean area and the contaminated area to ensure that the action of inserting the tip, filling the sample, and discarding the tip is a one-way flow. Each time the product changes from frozen to thawed, shake it appropriately or use a pipette to aspirate to ensure that the protein is evenly distributed within the system. This product is a native basement membrane matrix extracted from mouse tumors rich in extracellular matrix proteins. The main cytokines were Laminin, Col-IV, Entactin, Heparan sulphate proteoglycans and a variety of cytokines, such as insulin growth factor (IGF-1), transforming growth factor β (TGF-β), vascular endothelial growth factor (VEGF), epidermal growth factor (EGF), Fibroblast growth factor (bFGF), etc. The product is dissolved in high-glucose phenol red–free DMEM, and 50 μg/mL gentamicin was added to the non-customized product. By further screening on top of the low growth factor Matrigel, we have successfully optimized the organoid Matrigel to make it more suitable for organoid culture. Quality control: According to GB 14922.2-2022, the detection of viruses, pathogenic bacteria, parasites and bacteria in mouse populations was negative. The direct inoculation method was used to detect whether the product contained fungi and bacteria, and the result was negative. Extensive PCR testing for a wide range of pathogens, including LDEV, ensures strict control of the raw materials used in the production process. The mycoplasma sequence in the product was amplified using PCR technology, and the results were negative. Protein concentrations were determined using the BCA method. Endotoxin levels in products are measured using a gel limit check. After diluting the product in a ratio of 1:2 to the medium, the gel was placed at 37℃ for 30 minutes, and then the medium was added, and the product could maintain this form at 37℃ for 5 days. Stable gels can be formed by diluting the product to 70% content, adding 50 μL dropwise to a cell culture plate, and gelling at 37℃ for 30 minutes can form a stable gel, and after adding the medium, this form can be maintained in a 37℃ incubator for 15 days. Every batch of the product can undergo tumor cell invasion experiments and in vitro angiogenesis assays. Gelling time: 5-30min gel at room temperature, and the gelatinization speed is accelerated at 37℃
