1. Ensure reagents are fully thawed before use. Gently invert the tubes to mix contents thoroughly. Avoid creating bubbles during mixing. 2. Testing confirms this product remains effective after 10 freeze-thaw cycles. However，repeated freeze-thaw cycles should be minimized as they may degrade product performance. 3. Isothermal amplification is an ultra-high-sensitivity assay. Conduct testing in a standard PCR laboratory whenever possible. The isothermal amplification reaction setup area must be kept free from any potential contamination by amplification products. Although this kit employs UDG enzyme contamination prevention technology，do not open PCR tube caps. Seal isothermal amplification products and handle them according to post-amplification requirements to prevent ultra-high concentrations of amplification products from contaminating the experimental environment via aerosols or other means. 4. This product contains a color indicator. If the color differs significantly from the negative or positive controls after sample addition but before reaction initiation，dilute the sample，process it using a DNA extraction kit，or adjust the sample pH to near neutrality. 5. This isothermal amplification system requires high concentrations of divalent ions. Samples must not contain high concentrations of metal ion chelators such as EDTA. 6. We recommend using filter-tipped pipette tips to prepare the isothermal amplification system to minimize false positives caused by contamination. Product Description: Mycoplasma Colorimetric LAMP Assay Kit is a kit that amplifies mycoplasma DNA using loop-mediated isothermal amplification (LAMP) and detects mycoplasma contamination in cultured cells and other biological materials through a color change in the reaction system. This kit utilizes dU incorporation and UDG enzyme technology to prevent contamination of isothermal amplification products，allowing for rapid，effective，and highly sensitive detection of mycoplasma contamination. It also employs a visual color change technology，eliminating the need for electrophoresis; results can be determined by simply observing the color change with the naked eye. Product Features: 1. High sensitivity and short reaction time. This kit has a sensitivity 2-5 orders of magnitude higher than traditional PCR methods，with a detection limit of approximately 20-200 copies/μl calculated using a positive control plasmid. The detection reaction can be completed in just one hour. 2. High specificity and broad applicability to various mycoplasma species. This kit uses multiple specific LAMP primers designed based on the conserved 16S rRNA genomic sequence of mycoplasma，allowing for direct detection using cell culture supernatant or serum as a template. This kit can amplify approximately 45 types of mycoplasma，including those commonly found in cell culture contamination. This kit only amplifies mycoplasma DNA and does not amplify DNA from bacteria，fungi，or eukaryotic cells. 3. Visual color change technology eliminates the need for electrophoresis; results can be determined by simply observing the color change with the naked eye. This kit uses optimized indicator dyes. After rapid and extensive amplification of mycoplasma DNA fragments，a color change from violet or bluish-purple to sky blue or dark sky blue in the reaction tube indicates a positive result for mycoplasma. 4. Anti-contamination technology effectively prevents contamination. This kit uses dU and heat-labile UDG enzyme，which incorporates dU into the amplification products. Incubation at 30°C for 5-15 minutes before isothermal amplification effectively eliminates product contamination during the isothermal amplification process. During isothermal amplification，the heat-labile UDG enzyme is inactivated，thus not interfering with subsequent isothermal amplification detection. 5. This kit can directly detect samples suspected of mycoplasma contamination without the need for separate DNA extraction.
