Mouse Hepatic Kupffer Cell Isolation and Culture Kit

(1) You need to prepare the following materials: dissection board, 0.25% trypsin digestion solution, ice tray/ice box, surgical instruments (including 2 ophthalmic scissors, 2 straight surgical forceps, and 1 curved surgical forceps), tissue processing dishes (glass dishes, either 6 cm or 10 cm in diameter), 6 cm culture dishes, and several 15 mL/50 mL centrifuge tubes; (2) Before the formal experiment, it is recommended to use 1-2 normal mice for anatomical simulation training to familiarize yourself with the operational procedures and improve tissue separation efficiency; (3) Throughout the entire tissue separation process, it is advisable to operate on ice, but be careful not to make the tissue or liquid freeze; (4) The culture medium contains nutrients required for microbial growth. Please open it in a clean bench and aliquot, sealing the bottle opening with sealing film. Quick open and quick use, to avoid contamination; (5) The separation solution in this kit may exhibit crystal particle precipitation at the bottle opening or become turbid during storage, which is a normal phenomenon and can be used with no worries. This kit is specifically designed for the isolation of primary mouse hepatic Kupffer cells. It employs a mix of enzymes for low-temperature digestion, followed by density gradient separation and differential adhesion methods to purify mouse hepatic Kupffer cells. Laboratory validation has confirmed that each test (1T) can isolate 1×10? target cells. Immunofluorescence analysis confirms that the cell purity (CD68 positivity rate) exceeds 90%.
