(1) Before the formal experiment, it is recommended to use 1-2 normal mice for anatomical simulation training to familiarize yourself with the operational procedures and improve tissue separation efficiency; (2) Throughout the entire tissue separation process, it is advisable to operate on ice. However, avoid freezing due to excessive cold; (3) Reagent preparation or aliquoting must strictly adhere to aseptic technique guidelines. After aliquoting, immediately seal the containers with sealing film, use immediately upon removal, and avoid repeated freeze-thaw cycles or contamination. (4) The separation solution in this kit may exhibit crystal particle precipitation at the bottle opening or become turbid during storage, which is a normal phenomenon and can be used with no worries. This product is specifically designed for the isolation of primary mouse heart microvascular endothelial cells. It employs a two-step digestion method to digest tissue and density gradient centrifugation to purify cells. Laboratory validation has confirmed that it can isolate over 1×10? cells, with an immunofluorescence identification rate (CD31 positivity rate) exceeding 90%.
