Mitochondrial Membrane Potential and Apoptosis Detection Kit

This Mitochondrial Membrane Potential and Apoptosis Detection Kit is a dual-color fluorescence staining kit that combines Mito-Tracker Red CMXRos (also known as MitoTracker Red CMXRos), a red fluorescent probe dependent on mitochondrial membrane potential, and Annexin V-FITC, a green fluorescent probe for cell apoptosis, to simultaneously detect mitochondrial membrane potential and cell apoptosis in cultured cells. Red fluorescence labels viable cells with intact mitochondrial membrane potential, while green fluorescence marks apoptotic or necrotic cells. The kit is also supplemented with Hoechst 33342 nuclear fluorescent stain, which allows morphological observation of apoptosis by detecting nuclear fragmentation and chromatin condensation when needed. Featuring simple operation and clear results, this kit concurrently monitors two key apoptotic indicators: changes in mitochondrial membrane potential and phosphatidylserine externalization. After staining: viable cells show no green fluorescence but strong red fluorescence; apoptotic cells exhibit positive green fluorescence with markedly diminished or absent red fluorescence. The distinct red-green fluorescence differentiation enables detection via fluorescence microscopes, flow cytometers and other fluorescence analytical devices. It should be noted that the red fluorescence signal gradually fades upon the onset of apoptosis or necrosis, yet some cells with attenuated red fluorescence may not stain positive for green fluorescence. Additionally, phosphatidylserine externalization is an early event of apoptosis, and typical nuclear morphological alterations such as nuclear fragmentation and chromatin condensation are usually absent in the early apoptotic stage.
