Solarbio ELISA Kit uses the competitive enzyme immunoassay technique. A MEL antigen has been pre-coated onto a microplate. Add gradient-diluted standards and pre-diluted samples separately then add an antibody working solution and HRP-conjugated antibody working solution. MEL in the samples/standards competes with the plate-coated MEL for binding to the MEL antibody. After extensive washing a tetramethyl-benzidine (TMB) reagent is added to the wells for signal development. A Solution containing sulfuric acid is used to stop color development. The color intensity inversely proportional to the concentration of MEL is then measurable at 450nm. You can calculate the concentration of MEL in the samples by comparing the O.D. of the samples to the standard curve.
