1. Mycoplasma Detection Reagent A contains luciferase,which gradually inactivates with repeated freeze-thaw cycles. We recommend aliquoting for use,but ensure aliquoting containers are free from ATP contamination. Minor precipitation may occur in the reagent during repeated freeze-thaw cycles. In such cases,allow the reagent to equilibrate to room temperature and dissolve the precipitate as much as possible. If insoluble residue remains,centrifuge to remove it before use. Testing confirms this does not affect subsequent detection results. 2. Avoid repeated freeze-thaw cycles for the positive control; aliquoting for storage is acceptable. 3. Strongly recommend using white 96-well plates for testing. Transparent plates may cause interference between adjacent wells. 4. When testing cells,culture them normally for at least 24 hours (3-6 days is optimal). Use the supernatant from cell culture medium without trypsin digestion as the sample. For samples with extremely low mycoplasma content (e. g. ,serum,trypsin,antibiotics,or culture medium used in cell culture),testing after 2-3 days of culture is recommended. Insufficient incubation time may prevent detection of mycoplasma contamination. If extended incubation is impractical,consider using the Mycoplasma PCR Detection Kit (CA1081) or alternative methods. 5. For cell culture supernatants containing residual cells (e. g. ,adherent cell cultures) or suspension cell cultures,centrifuge at 200×g for 5 minutes at room temperature before collecting the supernatant for testing. Failure to do so may result in elevated background values. 6. Perform testing at approximately 20-25°C. Temperature affects both the chemiluminescence reaction and mycoplasma-specific enzyme-catalyzed reactions. Ensure both the detection reagent and samples are equilibrated to room temperature before testing. 7. Human skin surfaces contain abundant ATP. Wear gloves during testing to prevent reagent contamination. All other consumables must be clean and free of contamination. 8. This product is strictly for scientific research by qualified professionals only. It must not be used for clinical diagnosis or treatment,applied to food or pharmaceuticals,or stored in ordinary residential premises. Detection principle: The luminescent Mycoplasma Detection Kit utilizes the mycoplasma-specific enzymatic activity to detect contamination through an ATP-dependent luciferase-catalyzed luminescence reaction. By comparing ATP levels before and after adding the detection reagent,it determines whether samples are contaminated with mycoplasma. This kit enables rapid,effective,and highly sensitive detection of mycoplasma contamination. During cell culture,a small volume of culture supernatant can be sampled at any time to monitor for mycoplasma contamination.
