1. The detection reagents in this kit contain luciferase,which may gradually lose activity due to repeated freeze-thaw cycles. Although testing shows that freezing and thawing the reagent five times does not significantly affect its detection performance,for optimal results,it is recommended to aliquot and store the reagent after the first thaw. Ensure that the aliquoting containers are free from ATP contamination. During repeated freeze-thaw cycles,minor precipitation may occur in the detection reagents. In such cases,allow the reagents to equilibrate to room temperature and dissolve the precipitate as thoroughly as possible. If insoluble residues remain,centrifuge to remove them before use. Testing indicates this does not significantly affect subsequent detection performance. 2. Since luciferase activity is highly temperature-sensitive,both microbial samples and detection reagents must be equilibrated to room temperature before testing. Dissolve and thoroughly mix the detection reagent at room temperature or in a water bath not exceeding 25°C before use. Microorganisms cultured in the microplate must also be fully equilibrated to room temperature. Failure to do so may create a temperature gradient between the central wells and peripheral wells of the microplate,thereby affecting the intensity and stability of the fluorescence signal. 3. Compounds with high solvent content may interfere with the luciferase reaction,affecting chemiluminescence signals. Solvent interference can be excluded by including a control well containing microbial culture medium with solvent. Testing indicates that DMSO concentrations below 2% in the final assay system do not affect the reaction. 4. Use white or black 96-well or 384-well plates suitable for microbial culture. Transparent standard plates may cause cross-interference between adjacent wells during chemiluminescence detection. 5. While the manual provides a method for testing ATP standards,such testing is typically unnecessary when assessing microbial cell viability.
