1. Since luciferase activity is highly temperature-sensitive，both cells and detection reagents must reach room temperature before measurement. The detection buffer should be thawed and mixed at room temperature not exceeding 25°C or in a water bath at 20-25°C before combining with the detection substrate to form the detection reagent. 2. Prepare fresh solutions as needed. Any unused detection reagent may be aliquoted for storage，but ensure aliquoting containers are free from ATP contamination. 3. High solvent content in the test drug may interfere with the luciferase reaction，affecting the chemiluminescence signal. Solvent interference can be excluded by including a control well containing cell culture medium with the same solvent. Testing indicates that DMSO content below 2% in the final reaction system does not affect the reaction. 4. Use white or black 96-well or 384-well plates for assays. Transparent plates may cause cross-well interference. 5. While the manual provides ATP standard verification methods，such verification is typically unnecessary for routine cell viability assays.
