LH2 peptide is a cell-penetrating, pH-responsive peptide dimer with the amino acid sequence LHHLCHLLHHLCHLAG. In weakly acidic environments (such as the tumor microenvironment), this peptide can enhance intracellular uptake by tumor cells due to the protonation of histidine residues (pKa approximately 6). The complex PTX-LH2, formed by conjugating LH2 peptide with the anticancer agent paclitaxel, exhibited a stronger tumor-suppressive effect in a subcutaneous breast tumor model than paclitaxel alone. LH2 peptide shows promise as a delivery vector in anticancer research.
