Solarbio ELISA kit uses the quantitative competitive enzyme immunoassay (competitive ELISA) technique. An human E2 antigen has been pre-coated onto a microplate; Add gradient-diluted standards and pre-diluted samples separately then add a HRP-conjugated antibody specific for human E2 to allow competitive binding between soluble human E2 in the samples/standards and the plate-coated human E2 for the limited antibody binding sites. After extensive washing a tetramethyl-benzidine (TMB) reagent is added to the wells for signal development. A Solution containing sulfuric acid is used to stop color development. The color intensity inversely proportional to the concentration of human E2 is then measurable at 450nm. You can calculate the concentration of human E2 in the samples by comparing the O.D. of the samples to the standard curve.
