Human Colonic Organoid Medium, Expansion

Aliquoted organoid media should be stored at -20℃ for two years to avoid repeated freeze-thaw cycles. The thawed medium can be stored at 4℃ and is recommended to be used within two weeks. Organoid media contain bacterial and fungal antibiotics. The organoid medium should be thawed in a four-degree freezer, the thawed medium should be mixed upside down and mixed thoroughly, and the medium should be aliquoted in a sterile biosafety cabinet or clean bench, 10ml aliquots are recommended. Store the aliquot medium at -20 ℃, take out the aliquot medium and thaw it before use. Human colon organoid medium is used to establish and culture adult stem cell-derived human colon organoids. The self-renewal of colonic epithelial cells is driven by the proliferation of crypt-located stem cells and their progenitor cells, so that human colon organoids show all their characteristics. Colon epithelium has a strong influence on structure, cell type composition, and self-renewal dynamics. The research prospects of human colon development and disease are promising, and human colon organoids may also be applied to the in vitro expansion of the colon epithelium in regenerative biology.
