HA ELISA Kit

Solarbio ELISA kit uses the quantitative sandwich enzyme immunoassay technique. A recombinant human Aggrecan has been pre-coated onto a microplate. Add gradient-diluted standards and pre-diluted samples separately and allow them to fully combine with the recombinant human Aggrecan. After washing away any unbound substances a biotin-conjugated recombinant human Aggrecan is added; this biotin-conjugated Aggrecan binds specifically to the Hyaluronan already bound to the plate. Following a wash to remove any unbound combination horseradish peroxidase (HRP)-conjugated Streptavidin is added to the wells. After extensive washing a tetramethyl-benzidine (TMB) reagent is added to the wells for signal development. A Solution containing sulfuric acid is used to stop color development. The color intensity proportional to the quantity of bound protein is then measurable at 450nm. You can calculate the concentration of Hyaluronan in the samples by comparing the O.D. of the samples to the standard curve.
