Cells produce energy by ingesting a wide variety of nutrients and by intracellular metabolism. The process of nutrient metabolism varies with the extracellular environment,cell state,and cell type. Recent studies have found that nutrient metabolism is closely related not only to energy production,but also to a wide variety of cellular regulatory mechanisms such as gene expression. Glucose is one of the most important nutrients,and the process of cellular uptake of glucose is important for studying and understanding cellular function. The main method for evaluating glucose uptake in cells is radioisotope tracing. However,due to the complexity of radioisotope tracing,the versatility of radioisotope tracing is not high. In addition,there is an enzymatic cycling method that uses 2-Deoxy-D-glucose (2-DG),which can be used for plate detection but cannot be used for fluorescence microscopy and flow cytometry. Therefore,the most recently commonly used method is by fluorescence detection of the glucose analogue 2-NBDG. However,2-NBDG also has the problem of weak fluorescence intensity and low sensitivity,and the 2-NGDG ingested by cells may leak out of the cells. The fluorescent glucose analogue used in this product,Glucose Uptake Probe-Blue,is a more sensitive glucose uptake capacity assay than 2-NBDG. In addition,the use of the Washing and Imaging (WI) Solution included in this kit can inhibit probe leakage from the cell,resulting in more reproducible experimental data.
