Solarbio ELISA kit uses the competitive enzyme immunoassay technique.The D-Biotin coupled with BSA was coated on the microplate. Add gradient-diluted standards and pre-diluted samples separately and HRP-conjugated streptavidin was added. The biotin in the samples or standards competed with the coated biotin for the binding site on the HRP-conjugated streptavidin. After extensive washing a tetramethyl-benzidine (TMB) reagent is added to the wells for signal development. A Solution containing sulfuric acid is used to stop color development. The color intensity inversely proportional to the concentration of D-Biotin is then measurable at 450nm. You can calculate the concentration of D-Biotin in the samples by comparing the O.D. of the samples to the standard curve.
