Cholinesterase (ChE) Activity Fluorometric Assay Kit

Introduction: Cholinesterase (ChE, EC 3.1.1.8) is a group of enzymes that catalyze the hydrolysis of acyl-choline. It exists in various isoenzyme forms and is widely distributed in various tissues, serum, and body fluids of animals. It participates in the regulation of neural signals, lipid metabolism, and the maintenance of physiological internal environment homeostasis in the body. It is an important enzyme system in the body that undertakes the functions of substance hydrolysis and physiological regulation. Method: ChE catalyzes the hydrolysis of acetylcholine to produce choline. Choline is catalyzed by choline oxidase to generate H2O2. H2O2 reacts with a fluorescent probe under the action of peroxidaseto form a strong fluorescent product. The fluorescence intensity of this product at the excitation wavelength of 535 nm and the emission wavelength of 587 nm is proportional to the content of H2O2. This kit calculates the ChE activity by measuring the amount of H2O2 generated. Reagents and Equipment Required but Not Provided: Fluorescent microplate reader, balance, benchtop centrifuge, water bath/thermosetting incubator/metal bath, adjustable pipette, 96-well black microplate (fully black, 1 piece included in the box), sealing film (2 sheets included in the box), mortar/lymphomizer/cell ultrasonic disruption instrument, ice and distilled water.
