γ-Aminobutyrate transaminase (GABA-T ， EC 2.6.1.96) is a pivotal enzyme in the γ-aminobutyric acid (GABA) pathway ， catalyzing GABA’s degradation. GABA plays a fundamental role in the central nervous system by maintaining emotional stability ， promoting sleep ， regulating movement ， and preventing epilepsy ， primarily through its inhibitory function. Consequently ， GABA-T serves as a critical regulator of neuronal excitability and a key determinant of GABA concentration. In brain tissue ， the GABA-T catalyzed reaction utilizes α-ketoglutarate as an amino-group acceptor ， converting GABA into succinic semialdehyde and glutamate. Subsequently ， succinic semialdehyde dehydrogenase oxidizes succinic semialdehyde to succinate ， reducing NAD? to NADH in the process. This reduction forms the basis for the activity assay: NADH has a characteristic absorption peak at 340 nm ， and the rate of its increase is directly proportional to GABA-T activity.
