Braftide is a BRAF kinase allosteric inhibitor that acts on the surface of the BRAF kinase dimer, inhibiting its formation. The IC50 values of braftide against wild-type BRAF and oncogenic BRAFG469A are 364 nM and 172 nM, respectively. Upon binding to the TAT sequence, braftide inhibits the MAPK signaling pathway, preventing the proliferation of KRAS-mutant tumor cells (EC50 values of 7.1 μM and 6.6 μM for HCT116 and HCT-15, respectively).
