1.Storage conditions of each component: Staining solution should be stored at -20°C away from light; the inhibitor can be stored at room temperature or 4°C. 2.Avoid repeated freeze-thaw cycles of the staining solution，as repeated freezing and thawing will affect the experimental results. 3.For long-term storage，aliquot the staining solution into small portions according to usage requirements and store at -20°C. 4.Please use up the reagent as soon as possible after opening. Solarbio Anoikis Assay Kit is used to detect cell death caused by anoikis in living cells via a fluorescence method. The detection can be performed using a fluorescence microplate reader，fluorescence microscope，laser confocal microscope，or other fluorescence detection equipment with corresponding wavelengths. This kit contains a cell adhesion inhibitor. After being coated on cell culture vessels，the inhibitor can prevent cell adhesion. Living cells are detected by green fluorescence (excitation wavelength: 488 nm，emission wavelength: 516 nm)，while dead cells are detected by red fluorescence (excitation wavelength: 488 nm & 545 nm，emission wavelength: 617 nm). Anoikis is a type of programmed cell death and a special form of apoptosis. Normal adherent cells maintain intercellular connections and tightly adhere to the extracellular matrix (ECM) to form a survival niche. Once detached from the niche，the cells will undergo apoptosis. The adhesion of adherent cells to the extracellular matrix (ECM) is of great significance for their growth and proliferation. As a special form of programmed cell death，anoikis induced by the loss of normal adhesion or abnormal adhesion to the ECM plays a vital role in organism development，tissue homeostasis，disease pathogenesis and tumor metastasis. The adhesion between cells and the extracellular matrix (ECM) is critical for cell survival.
