β-hydroxybutyrate dehydrogenase ( β-HBDH;EC1.1.1.30 ) is a key enzyme in the process of ketone body redox reaction. Ketone bodies are an important marker for detecting metabolic control, so accurate detection of β-hydroxybutyrate dehydrogenase activity is critical. At pH 7.0 and 37 °C, acetoacetate is reduced to 3-hydroxybutyrate or decarboxylated to acetone by β-HBDH, and NADH is oxidized to NAD +. NADH has a characteristic absorption peak at 340 nm. The activity of β-HBDH can be calculated by detecting the change of absorbance at 340 nm.
